The viral genome mutates through random replication errors, just like any DNA. Antigenic drift is caused by exactly that.
There is no transcriptase, just a reverse transcriptase. What you mean is called RNA polymerase.
Protease inhibitors do not primarily block replication and Ivermectin is not primarily a protease inhibitor.
In a titer test for Covid you measure viral particles/genomes, not a specific protein.
