If you're talking about speeds above 30 mph in a truck with aftermarket parts (i.e. lift kit) then it's likely that your front driveshaft does not have the proper angles to run smoothly.
I just read your question again and the above may not specifically apply; however, when the hubs are locked the driveshaft is loaded and will have different "vibration" characteristics.
What speeds are you talking about? Are you talking about on dirt or pavement or both?
If you have a locker you could try locking one hub at a time to see what happens.
