The 1uzfe (4.0L)and the 2uzfe (4.7) have the same block configuration, but the 2uzfe is cast iron.  Obviously, the internals are different, but they both have 6 bolt main bearings.  From what I understand, the transmissions can be swapped between them.  The 1uzfe had 3 different oil pan configurations : front sump, mid sump and rear sump, all of which will swap onto the 2uzfe.  The 2uzfe 2wd and 4wd tundras had the same oil pan configuration, but the Landcruisers have a different configuration (not sure what).  The 1uzfe also had different exhaust manifolds - 2, mabye 3 different configurations due to the 3 different body styles in came in.  I beleive the heads are somewhat different between the 1uzfe and the 2uzfe - at least the gs400 heads are different (high flow).  The sc400's had a hydraulic cooling fan ran off a pulley/ pump configuration when the ls400's and the gs400's did not.   Oh, The intakes are different too.  The 1uzfe has a side mount throttle body on most of the older ls400's and sc400's and around '98 or '99 they went to a front mount throttle body with a VVTi setup.  I beleive around 2001, the 1uzfe got bumped up to a 4.3L, hence the name LS430, SC430, GS430 - I beleive this was the 3uzfe.  The 2uzfe got a change in '05 and went to a composite intake and a vvti setup.  Sometime in '04, toyota changed the trans in the Tundras and now they don't have a dipstick and run WS trans fluid that's supposed to get changed every 100k miles.  The 2uzfe has always had a direct ignition system but the 1uzfe started of with a dual coil pack system with a rotor and distributor cap on each cam sprocket and eventually went to a direct ignition setup.  The early versions of the 1uzfe engine had an EGR system and eventually Lexus got that engine to burn clean enough, so they removed it from production.  The 1uzfe never had an egr system.  Some of the early LS400's had a air injection system too ('92-93 I believe).

Hope this helps,  I'm nt positive on all my info, I'm just trying to remember from my days working at Lexus and now working for Toyota.
